Overview
Retatrutide is a novel triple incretin receptor agonist (GLP-1, GIP, and glucagon) developed by Eli Lilly. In Phase 2 clinical trials it demonstrated the highest percentage weight loss of any single therapeutic agent studied to date, with Phase 3 trials ongoing as of 2024โ2025.
Mechanism of Action
Retatrutide activates three receptors simultaneously: GLP-1 receptors to reduce appetite and improve glycemic control; GIP receptors to augment insulin secretion and lipid metabolism; and glucagon receptors to increase energy expenditure and hepatic fat oxidation. This triple agonism produces synergistic and profound metabolic effects.
